Skrining Aktinomisetes Pendegradasi Nikotin Pada Daun Tembakau (Nicotiana tabacum L.)

Kabupaten Jember merupakan salah satu sentral penghasil tembakau tertinggi di Jawa Timur dan pada tahun 2018 produksinya mencapai 163,267,5 ton. Produksi tanaman menghasilkan limbah organik yang berbahaya yaitu nikotin. Nikotin alkaloid beracun aktif, berminyak, tersusun atas unsur karbon, hidrogen, nitrogen, sangat larut terhadap alkohol, eter, minyak tanah, air. Kelarutannya berisiko mengalami leaching (pencucian) selama masa penyimpanan tembakau. Hal tersebut mengakibatkan nikotin tercuci dapat mengalir ke dalam badan air sehingga membahayakan makhluk hidup lingkungan. Beberapa mikroorganisme mampu mendegradasi dengan memanfaatkannya sebagai sumber karbon nitrogen untuk pertumbuhannya, satunya aktinomisetes. Penelitian ini bertujuan mendapatkan aktinomisetes asal rhizosfer memiliki aktivitas pendegradasi besar daya degradasinya. dilakukan skrining 23 isolat indikator tumbuh media hingga terdapat 3 terpilih, uji degradasi cair metode titrasi daun Balai Pengujian Sertifikasi Mutu Barang (BPSMB) kota Surakarta, identifikasi morfologi secara mikroskopis terpilih menggunakan cover slide method. Hasil menunjukkan bahwa 5 kemampuan tumbuh, 13 mumpu baik, lebih baik 2 tidak tumbuh. Isolat ATG 60, 68, 69 (NIM). Daya 60 (2,33% 30,56%); 68 (2,53% 31,16%); (2,1% 25,82%). Identifikasi struktur spora masing-masing adalah Saccharopolyspora sedangkan genus Streptomyces.
